Phenol-sulfuric acid assay
WebFeb 26, 2024 · Here, we developed a phenol-sulfuric acid (PSA) assay in a microplate format to quantify soluble sugars in ruminant feeds. This method is easy and quick and requires only a small quantity of harmful reagents. We found that assay measurements were not affected by the representative organic acids and sugar alcohol contained in feeds. WebThe objective of this lab was to have students use a spectrophotometer to analyze the carbohydrate content in the sample. Phenol-sulfuric acid methods were also applied to determine the total quantity of carbohydrates in the food. It is important to study carbohydrate content because the absorbtivity of different carbohydrates varies.

WebCarbohydrates form either furfuraldehyde or its homologs with strong acid. These derivatives of the carbohydrates produce colored compounds by polymerization or condensation with aromatic phenols. This is the basis of the Molisch reaction and also of Bial’s orcinol assay (1902) which was originally designed for pentoses and hexuronic acids. WebPrinciple Total carbohydrate concentration is based on the phenol-sulfuric acid method in which polysaccharides are hydrolyzed and then converted to furfural or hydroxylfurfural. …
WebIn the assay, polysaccharides (mono, di, tri, etc.) and their derivatives, in the presence of sulfuric acid, are hydrolyzed to monomers and converted to furfural or hydroxyfurfural. These react with the developer to form a chromogen that can be quantified by measuring the absorbance at λ = 490 nm.
